I've been studying a lot  how to do a non-invasive melanocyte stem cell transplantation (at home or at your family doctor clinic).  It took me 4 months and 3 very thick books, to get to a step-by-step procedure, that has to be shared with a Doctor.  Everything has to be sterilized.

The good thing is that the whole procedure takes about 2 hours.  The procedure can be repeated as many times as you want, respecting a proper recovery interval.  The total investment is around US$ 300, for 10 procedures.  No air ticket, not hotel, etc.

I do believe it's worth a try, and I will in a close future.

Procedure:

1- Pluck the hair with a forceps or a good tweezer. 20 hair follicles are enough for 1cm2 and put them on a petri #1.  Hair bulb have to come together (as intact as possible).

2- Add 10ml of 0.025% trypsin-EDTA solution to petri #1.  Use a dropper.

4- Transfer the hair follicles to another petri #2.

5- Add 5ml of trypsin inhibitor to petr #1 to avoid melanocyte digestion.

6- Keep petri #1 separated apart (waiting).


7- Add 10ml of 0.025% trypsin-EDTA to petri#2.


8- Put petr #2 in a incubator for 30 min.


9- Transfer the hair follicles to another petri #3.

10- Add 5ml of trypsin inhibitor to petr #2 to avoid melanocyte digestion.


11- Keep petri #2 separated apart (waiting).


12- Add 10ml of 0.025% trypsin-EDTA to petri#3


13- Put petr #3 in a incubator for 30 min. Add 5ml of trypsin inhibitor to petr #3 to avoid melanocyte digestion.


14- Throw the hair away and mix all petri  (#1, #2, and #3) contents.


15- Filter the combined solution with a 70um strainer.

16- Centrifuge the strained solution during 5min at 1000RPM, so that it will preciptate.

17- Remove the supernatant liquid and keep only the pellets .


18- After removing the supernatant liquid, add a 5-10ml of saline solution (PBS PH 7.4 or DMEM/F-12) to the pellets and mix it.

19- You've got an autologus stem melanocyte suspension.  Keep in the refrigerator.


20- Roll dermaroller (0.5mm needles) on the patches (both vertically and horizontally) twice,  where suspension will acommodate. Use a dropper.

21- Then two layers of saline moistened woven gauze compresses semipermeable Tegaderm™ film are applied and glued in place onto the surrounding skin with Mastisol® (Ferndale Laboratories, Inc, Ferndale, MI, USA). The glue ensures that it will be secured in place for the period needed.

22- Keep the dermarolled regions as stopped as possible during a week and also don't  water or moisten them for the same time.

    I went through this procedure thrice and it is effective, this is an OPD based procedure will take max 2-3 hours, and it is very effective for localized small patches, wherein for larger patches procedure has to be repeated depending upon the size. I went through this procedure on my foot ankles of both the legs (both sides) and the results were quite good although the entire patch has not been recovered fully, a small area is still effected, will undergo once again with the procedure and me and my doctor is very hopeful that this time the entire region will get recovered. Will update you guys once its done.
